Bis(2-oxo-3-oxazolidinyl)phosphinic Chloride (BOP-Cl) (CAS: 68641-49-6), reagents for carboxyl activation and peptide coupling. BOP-Cl was used in the preparation of hexadepsipeptide. Also used as reagent is used for activating the carboxylic group, synthesis of amides, esters and peptides. BOP-Cl is a highly effective reagent for the acylation of N-methyl amino acids. It provides good yields with low epimerization. BOP-Cl is also an efficient reagent for coupling protected amino monothioacids to iminoamino acid residues to form thiopeptide bonds.
BOP-Cl is a phosphoric acid derivative used as a coupling reagent for amide and ester formation. BOP-Cl is effective in the acylation of N-methylated amino acids and in the introduction of thiopeptide bonds on imino acids. BOP-Cl requires mild conditions, gives good yields and suppresses epimerisation. BOP-Cl and other phosphinic acids activate the carboxylic acid by forming a carboxylic-phosphinic mixed anhydride, in which stereochemistry is conveniently retained. BOP-Cl gives good performances in the acylation of N-methylated amino acids due to intramolecular base catalysis.
